@Zero Knievel had started a talk about this.  I had given the normal cautions about interactions, risks, and real potential dangers.  Ironically, in chasing ongoing health challenges, it was specifically recommended for me.  This is just part of it, which I have verified through three different sources.

 

Root:  I have a dual gene defect that affects the ability to use B vitamins, and the DOPA>Dopmamine>Serotonin chain.

 

Start with the methylation foundation (Pure TheraPro daily + Thorne 2–3×/wk) since you may already be sourcing those. Then layer in the BH4 cofactors — vitamin C and NMN are cheap and low-risk with immediate relevance. NA Semax Amidate is the highest-impact add on the peptide side and should be your first peptide given it directly addresses the BH4/neurotransmitter signaling deficit rather than just precursor-loading. BPC-157 is a natural second peptide to run concurrently or sequentially given the D2 receptor upregulation and protective effects.

 

Methylene blue specifically: The mechanism is genuinely useful for your situation — it acts as an alternative electron carrier that can directly reduce the BH3 radical back to active BH4, meaning it partly addresses the root deficiency rather than just compensating downstream. The real risk vector is the MAOI activity, which at 1–10 mg is mild but real. Since you don't use 5-HTP, your main exposure is L-tryptophan in the evening and SAMe in the morning. Structure it as: MB days = no tryptophan that evening, conservative on SAMe. Non-MB days = tryptophan and SAMe freely. The G6PD check is worth doing if you haven't — it's a standard item on a comprehensive blood panel or 23andMe data can tell you (rs1050828, rs1050829).

I started methylene blue also. I've been on it almost a month. I have noticed a difference in the neuropathy in my right foot I'm starting to get some of the feeling back but still have a lot of numb places. They want to fuse my lower spine but I am so freaked out by it.

Just do it.  A friend had this done, and she says she wishes she'd done it sooner.  For her it was near instant relief from years of pain.

 

Is the neuropathy related?  And if so I wonder how the MB can be helping?  Or is it just a local issue?

The neuropathy is from my spine issue. I think it's from the antinflammatory and antioxidant properties and helps with excessive nerve firing.

They want to do S1 up to T10.

I have a big deductible...It's a lot to process getting it done...

Sounds like continuing the MB and see what happens makes sense?

 

Also something to research:  GLOW and KLOW peptide blends.  GLOW is GHK-Cu, BPC-157, and TB4 or TB500.  KLOW also adds KPV.  All are correlated with rebuilding tissue and some of my friends have seen miracle results with major skeletal issues.  I've seen a good improvement of milder issues, but very noticeable, so I'll use it on and off.  It has one effect that can't be subjective and is clear evidence that it's doing something.  I've had a knee that makes noise going up stairs for decades.  When I'm on this, the noise stops.  When I'm off, after a couple months, it comes back.

 

Unlike the other peptides, this has a very short half life and results can be apparent in a couple weeks, or take 1-2 months.  One friend with debilitating knee problems was running up the stairs after ten days on it.
